目的研究子宫腺肌病中孕激素受体亚型的表达,探讨孕激素受体亚型(hPR)在子宫腺肌病发病中的作用机制。方法运用免疫蛋白印迹法(Western blot)和RT-PCR方法测定子宫腺肌病患者(24例)病灶及其在位内膜及对照组(19例)子宫内膜标本中孕激素受体亚型在蛋白和mRNA水平的表达,计算机图像分析、半定量研究孕激素受体亚型的表达。结果子宫腺肌病异位内膜中hPRs的表达均明显低于在位内膜和对照组,研究组在位或异位内膜中,hPR-A含量均高于hPR-B;而在mRNA水平上则差异无统计学意义(P>0.05)。结论子宫腺肌病异位内膜hPRs蛋白的表达均明显低于在位内膜,而在mRNA水平上却无显著性差异;hPR蛋白的活性可能受转录后调节。
Objective To study the expression of progesterone receptor subtypes in adenomyosis and explore the mechanism of progesterone receptor subtypes (hPR) in the pathogenesis of adenomyosis. Methods Western blot and RT-PCR were used to detect the expression of progesterone receptor subtype in endometriosis and endometriosis (19 cases) of patients with adenomyosis (24 cases) Expression at protein and mRNA levels, computer image analysis, semi-quantitative study of progesterone receptor subtype expression. Results The expression of hPRs in ectopic endometrium of adenomyosis was significantly lower than that of eutopic endometrium and control group. The content of hPR-A in eutopic or ectopic endometrium of study group was higher than that of hPR-B. The difference was not statistically significant (P> 0.05). Conclusions The expression of hPRs in ectopic endometrium of adenomyosis is significantly lower than that in eutopic endometrium, but not in mRNA level. The activity of hPR protein may be regulated by transcription.
