目的:在单因素实验的基础上,利用表面响应曲面法(RSM)对HPLC-UV法测定牛磺罗定含量中的牛磺罗定衍生化条件进行优化。方法:以衍生化试剂不同的浓度、反应时间和反应温度为因子,以反应后衍生化产物的量(用峰面积表示)为响应因子,对衍生化条件进行优化。结果:最佳衍生化条件是反应温度为100℃,衍生化试剂浓度为20 g.L-1,反应时间为88 min。结论:本方法结果准确可靠,可应用于牛磺罗定的衍生化。
OBJECTIVE: To optimize the derivatization conditions of taurolidocetin in taurolidine by HPLC-UV method based on single factor experiments and surface response surface methodology (RSM). Methods: The derivatization conditions were optimized by taking different concentrations of derivatization reagent, reaction time and reaction temperature as factors, and the amount of derivative product after reaction (expressed as peak area) as response factor. Results: The optimal derivatization conditions were as follows: the reaction temperature was 100 ℃, the concentration of derivatization reagent was 20 g.L-1 and the reaction time was 88 min. Conclusion: The method is accurate and reliable and can be applied to derivatization of taurolidine.
