目的研究补肾益气生精中药对去卵巢(OVX)大鼠骨组织中信号转导分子Smad2的表达,以及在骨质疏松症发病中的作用机制。方法用去卵巢的方法建立绝经后骨质疏松症模型,给予补肾益气生精中药防治12周,以尼尔雌醇为阳性对照,用双能-X线骨密度仪检测大鼠离体股骨骨密度,用逆转录多聚酶链反应(RT-PCR)、W estern blot方法分别检测Smad2的mRNA、蛋白表达。结果卵巢切除大鼠离体股骨骨密度显著下降,骨组织中Smad2的蛋白及mRNA表达水平均下降。补肾益气生精中药能够提高去卵巢大鼠的骨密度,并且可以从蛋白及mRNA水平提高骨组织中Smad2的表达。结论骨组织中信号转导分子Smad2表达水平的下调可能是原发性骨质疏松症发生的重要机制,补肾益气生精中药通过上调Smad2的表达而有效改善骨密度。
Objective To investigate the expression of Smad2, a signal transduction molecule in bone tissue of ovariectomized (OVX) rats, and its mechanism in the pathogenesis of osteoporosis. Methods The postmenopausal osteoporosis model was established by ovariectomy. The kidney was treated with Bushen Yiqi Shengjing for 12 weeks. Nilestriol was used as positive control and the rat femur was detected by dual energy X-ray absorptiometry. Bone density was measured by reverse transcription-polymerase chain reaction (RT-PCR) and W estern blot method respectively to detect mRNA and protein expression of Smad2. RESULTS: Bone mineral density of femurs in ovariectomized rats was significantly decreased, and protein and mRNA expression of Smad2 in bone tissue decreased. Bushenyiqishengjing can increase bone density in ovariectomized rats, and can increase the expression of Smad2 in bone tissue from protein and mRNA levels. Conclusion The down-regulation of signal transduction molecule Smad2 in bone tissue may be an important mechanism of primary osteoporosis. Bushen Yiqi Shengjing herbs can effectively improve bone mineral density by up-regulating the expression of Smad2.
