目的:探讨波形蛋白在子痫前期蜕膜组织中的表达及意义。方法:采用荧光实时定量PCR(SYBR GREEN I assay)、Western blotting和免疫组织化学技术分别对晚发型重度子痫前期(A组)及早发型重度子痫前期(B组)和正常足月妊娠妇女(对照组,C组)各10例的波形蛋白mRNA及蛋白进行测定。结果:波形蛋白主要表达于蜕膜细胞的胞膜、胞浆及间质。荧光实时定量PCR,Western blotting和免疫组化表达等各方法验证结果显示:波形蛋白mRNA及蛋白在C组蜕膜组织中表达最高;B组中次之;A组中最低,各组间差异有统计学意义(P<0.05)。结论:波形蛋白表达下调可能参与子痫前期的病理过程,并且波形蛋白下调的程度同子痫前期发病的早晚有关。
Objective: To investigate the expression and significance of vimentin in decidual tissue of preeclampsia. Methods: The serum levels of IL-10 and IL-6 in late-onset severe preeclampsia (group A), early-onset severe preeclampsia (group B) and normal term pregnant women Control group, group C) 10 cases of vimentin mRNA and protein were measured. Results: The vimentin was mainly expressed in the membrane, cytoplasm and interstitium of decidual cells. Fluorescence real-time PCR, Western blotting and immunohistochemistry were used to verify the results. The results showed that the expression of vimentin mRNA and protein was the highest in group C, the group B was the second, the group A was the lowest, and the difference among groups was Statistical significance (P <0.05). Conclusion: The down-regulation of vimentin may be involved in the pathological process of preeclampsia, and the extent of down-regulation of vimentin is related to the onset of preeclampsia.
