目的:APOBEC3B(A3B)是载脂蛋白B mRNA编辑酶催化多肽(APOBEC)家族的重要成员,本文旨在探讨其与非小细胞肺癌(NSCLC)预后及顺铂耐受的关系。方法:实时荧光定量PCR(qRT-PCR)分析40例NSCLC组织中A3B mRNA的表达,Kaplan Meier plotter分析A3B表达与临床预后的相关性,并在人肺腺癌细胞A549中构建A3B低表达细胞系,通过MTS及平板克隆实验观察细胞顺铂敏感性的改变,γ-H2AX免疫荧光定量检测DNA损伤。结果:与癌旁组织比较,A3B在NSCLC组织中高表达(27/40),Kaplan Meier plotter分析显示A3B表达与NSCLC总生存率(OS)呈正相关[腺癌:n HR=0.64(0.47-0.86),n P=0.002 6;鳞癌:n HR=0.77(0.59-1.01),n P=0.006]。体外实验发现,敲低表达A3B后细胞对顺铂耐药性增强,DNA损伤修复增加。n 结论:A3B介导了肺癌对顺铂的敏感性,这一作用可能部分解释A3B高表达的NSCLC患者有更好的预后。“,”Objective:APOBEC3B (A3B) is an important member of the apolipoprotein B mRNA editing enzyme catalytic polypeptide (APOBEC) family. The study aims to investigate the relationship between A3B expression and prognosis as well as resistance to cisplatin in non-small cell lung cancer (NSCLC).Methods:Real-time quantitative polymerase chain reaction (qRT-PCR) was used to analysis the A3B mRNA expression in 40 NSCLC tissues; Kaplan Meier plotter was used to analyse the correlation between A3B expression and clinical prognosis; in addition, the knock-down A3B expression cell line in human lung adenocarcinoma cell A549 was constructed; MTS and plate cloning experiment were performed to observe the changes in cell cisplatin sensitivity, and γ-H2AX immunofluorescence was used to quantitate the DNA damage.Results:Compared with adjacent tissues, A3B was highly expressed in NSCLC tissues (27/40). Kaplan Meier plotter analysis showed that A3B expression was positively correlated with NSCLC overall survival (OS) [adenocarcinoma: n HR=0.64(0.47-0.86), n P=0.002 6; squamous cell carcinoma: n HR=0.77(0.59-1.01), n P=0.006]. Cell-based studies showed that the knock-down A3B expression contributed to sensitivity to cisplatin in A549 cells.n Conclusions:A3B mediates the sensitivity of lung cancer to cisplatin. This effect may partly explain why NSCLC patients with high A3B expression have a better prognosis.
