目的:研究秃疮花中提取异紫堇碱的最佳工艺条件及采用大孔吸附树脂分离纯化异紫堇碱的方法。方法:采用正交试验设计,优化异紫堇碱的提取工艺;通过静态吸附/解吸附试验筛选最佳吸附异紫堇碱的大孔树脂。结果:最佳提取条件为:料液比1∶15(g/mL)、1%的酸水溶液为溶剂,提取时间1h×3次;LX28树脂为最佳吸附树脂。结论:采用最佳试验条件提取秃疮花,异紫堇碱得率可达0.88%;提取液经LX28树脂分离纯化,得到质量分数为85.34%的异紫堇碱,产率达68.64%。
Objective: To study the optimal conditions for the extraction of corydaline in bald flower and the method of separating and purifying corydaline with macroporous adsorption resin. Methods: Orthogonal experiment design was used to optimize the extraction process of corydaline. The best adsorption of corynebacterium macroporous resin was screened by static adsorption / desorption test. Results: The optimal extraction conditions were as follows: solid-liquid ratio 1:15 (g / mL), 1% aqueous acid solution as solvent and extraction time 1h × 3 times. LX28 resin was the best adsorption resin. CONCLUSION: The optimal extraction conditions were as follows: the rate of corynebacterium was 0.88%; the extract was separated and purified by LX28 resin to obtain the corynebacterium with the mass fraction of 85.34%, the yield was 68.64%.
