目的检测miR-1246、ZO-1、vimentin在乳腺正常上皮细胞MCF10A和乳腺癌癌细胞MCF7、MDA-MB-231中的表达,并探讨miR-1246在促进乳腺癌上皮-间质化(epithelial-mesenchymal transition,EMT)中的作用。方法用荧光定量PCR法检测乳腺正常上皮细胞MCF10A、乳腺癌细胞MCF7、MDA-MB-231中miR-1246的相对表达量,用免疫印迹法检测乳腺正常上皮细胞MCF10A、乳腺癌细胞MCF7、MDA-MB-231中紧密连接蛋白ZO-1、波形蛋白vimentin的表达情况。采用SPSS 16.0统计软件分析,用U检验、t检验和Spearman相关分析分析数据。结果miR-1246和ZO-1在恶性乳腺癌细胞中的表达明显降低(P<0.05),vimentin在恶性乳腺癌细胞中的表达明显增加(P<0.05),miR-1246与ZO-1表达呈正相关(r=0.940,P<0.05),miR-1246与vimentin表达呈负相关(r=-0.714,P<0.05)。结论 miR-1246表达水平与EMT标志物ZO-1、vimentin的表达水平存在一定的相关性,提示miR-1246在促进乳腺癌EMT过程中扮演重要角色。
Objective To detect the expression of miR-1246, ZO-1 and vimentin in breast normal epithelial cells MCF10A and breast cancer cells MCF7 and MDA-MB-231 and to explore the role of miR-1246 in promoting epithelial- mesenchymal transition, EMT). Methods The relative expression of miR-1246 in normal breast epithelial cells MCF10A, breast cancer MCF7 and MDA-MB-231 cells was detected by real-time fluorescence quantitative PCR. The expression of miR-1246 in normal breast epithelial cells, MCF7 breast cancer cells and MDA- MB-231 in the tight junction protein ZO-1, vimentin vimentin expression. Using SPSS 16.0 statistical software analysis, U-test, t-test and Spearman correlation analysis of the data. Results The expression of miR-1246 and ZO-1 in malignant breast cancer cells was significantly decreased (P <0.05), and the expression of vimentin in malignant breast cancer cells was significantly increased (P <0.05). The expression of miR-1246 and ZO-1 was positive (R = 0.940, P <0.05). There was a negative correlation between miR-1246 and vimentin expression (r = -0.714, P <0.05). Conclusion The expression level of miR-1246 is correlated with the expression of EMT markers ZO-1 and vimentin, suggesting that miR-1246 plays an important role in promoting EMT in breast cancer.
