综合运用硅胶柱色谱、Sephadex LH-20、聚酰胺柱色谱、半制备HPLC以及重结晶等方法对毛果鱼藤中黄酮类化学成分进行分离纯化,并通过质谱、核磁共振谱等方法鉴定各单体化合物的结构。最后从毛果鱼藤的乙酸乙酯部位分离得到9个黄酮类化合物,依次为香叶木素(1),3,3’-二甲醚-槲皮素(2),阿弗洛莫生(3),6,3’-dihydroxy-7,4’-dimethoxyisoflavone(4),飞机草素(5),7,3’-dihydroxy-8,4’-dimethoxyisoflavone(6),6,4’-dihydroxy-7,3’-dimethoxyisoflavone(7),5,7,4’-trihydroxy-3,3’,5’-trimethoxyflavone(8),alpinumisoflavone(9)。所有化合物均为首次从该植物中分离得到。运用MTT法进行体外抗肿瘤实验,结果表明化合物2显示一定的抑制细胞增殖活性。
The flavonoids were separated and purified by silica gel column chromatography, Sephadex LH-20, polyamide column chromatography, semi-preparative HPLC and recrystallization, and identified by mass spectrometry and nuclear magnetic resonance spectroscopy Structure of the compound. All compounds were isolated from this plant for the first time. MTT assay in vitro antitumor experiments showed that the compound 2 showed a certain inhibition of cell proliferation activity.
