目的 观察羟基磷灰石(HA) - 软骨细胞复合物植入自体后软骨细胞的转归.方法以块状多孔HA为载体培养大鼠肋软骨细胞构建这种复合物,在同一大鼠的胫骨上段制作膜内化骨模型,将这种复合物分别植入模型的骨缺损处和臀部皮下组织内,对侧植入块状多孔HA 进行对照实验,于14 天和60 天取材,制作不脱钙组织切片,以苏木精- 亮绿- 藏红T 进行染色,软骨组织染成鲜红色,骨组织染成灰绿色.结果 骨缺损处(5 侧) 和皮下组织(5 侧) 实验组均有软骨组织和骨组织形成,对照侧则没有.结论 将HA- 软骨细胞复合物植入体内后,其中的软骨细胞能够存活并产生软骨组织和骨组织.“,”Objective To investigate the fate of chondrocytes in the hydroxyapatite(HA)-chondrocytes composite followng in vivo implantation.Methods The composite material was organized by cultivating chondrocytes obtained from costicartilage of rat in the pore of HA blocks for seven days, then was implanted in surgically created defect of upper tibia and in the subcutis of hip seperately in the same rat,the HA without chondrocytes served as a control. The specimens were harvested at 14 and 60 days postimplantation,then were processed by undecalcified technique.The serial sections were stained with light blue and safranin T to show the formation of bone and cartilage.Results At 14 day postimplantation,no cartilage and bone forming inside of the pores of the HA.At 60 day, in the experimental group,there are cartilage and bone formation inside of the pores of the HA implaned into both bone defects and subcutis but not the HA controls. Conclusion The chondrocytes in the pores of HA-chondrocytes composite were able to live,expand and form cartilage and bone through endochondral osteogenesis.
