目的研究磷酸化蛋白激酶B(pPKB)和缺氧诱导因子1α(HIF-1α)在喉鳞状细胞癌(LSCC)中的表达及其与LSCC的肿瘤分期、分化程度和局部淋巴结转移的关系。方法应用免疫组织化学SABC法,检测50例LSCC标本癌组织和20例声带息肉中pPKB和HIF-1α的表达情况。结果LSCC中pPKB和HIF-1α蛋白的阳性表达率显著高于声带息肉组(68.0%vs0,χ2=26.44,P<0.01;64.0%vs0,χ2=23.58,P<0.01)。LSCC中pPKB和HIF-1α蛋白的阳性表达率,在有无淋巴结转移、不同T分期、高分化与中低病理分化的LSCC中比较,差异均有统计学意义(P均<0.05)。结论pPKB和HIF-1α在喉癌组织中表达明显升高,pPKB和HIF-1α可能与喉癌的发生、浸润和转移密切相关,其联合检测可能成为LSCC早期诊断及判断恶性程度的指标之一。
Objective To investigate the expression of phosphorylated protein kinase B (pPKB) and hypoxia inducible factor 1α (HIF-1α) in laryngeal squamous cell carcinoma (LSCC) and its relationship with tumor stage, differentiation and local lymph node metastasis in LSCC. Methods Immunohistochemical SABC method was used to detect the expression of pPKB and HIF-1α in 50 cases of LSCC and 20 cases of vocal cord polyp. Results The positive rates of pPKB and HIF-1α in LSCC were significantly higher than those in vocal cord polyp (68.0% vs0, χ2 = 26.44, P <0.01; 64.0% vs0, χ2 = 23.58, P <0.01). The positive rates of pPKB and HIF-1α in LSCC were significantly different between LSCC with or without lymph node metastasis, different T stage, well differentiated and moderately low pathological differentiation (all P <0.05). Conclusions The expressions of pPKB and HIF-1α in laryngeal squamous cell carcinoma are obviously increased. The expression of pPKB and HIF-1α may be closely related to the occurrence, invasion and metastasis of laryngeal carcinoma. The combined detection of pPKB and HIF-1α may be one of the indicators of LSCC early diagnosis and malignancy .
