目的了解RhD抗原免疫刺激途径,建立D抗原免疫刺激活化的模型。方法选取RhD血型不同的供者,采用红细胞与单个核细胞直接混合培养,以及DC细胞负载RhD抗原后与单个核细胞混合培养2种实验方案,流式细胞仪检测CD4~+和CD8~+T细胞的活化量。结果 RhD(-)个体的DC细胞负载D抗原后,可以有效活化RhD-个体的CD4~+和CD8~+T细胞,而直接混合培养方式则不能。结论本研究采用DC细胞负载RhD抗原的方法,成功诱导了RhD(-)个体的T细胞活化和增殖,验证了RhD抗原的胸腺依赖性抗原特性,建立了D抗原免疫刺激T细胞活化的模型,可以用于输血免疫的研究。
Objective To understand the immunostimulatory pathway of RhD antigen and establish a model of immunostimulation activated by D antigen. Methods Two donors with RhD blood group were selected. Two kinds of experimental protocols were established by direct mixed culture of erythrocytes and mononuclear cells and mixed with mononuclear cells after DC cells were loaded with RhD antigen. Flow cytometry was used to detect CD4 ~ + and CD8 ~ + T The amount of cellular activation. Results RhD - (+) DCs could effectively activate RhD - stimulated CD4 ~ + and CD8 ~ + T cells after they were loaded with D antigen, but not in direct mixed culture. Conclusions In this study, DCs were loaded with RhD antigen and successfully induced activation and proliferation of T cells in RhD (-) individuals. The thymus-dependent antigenicity of RhD antigen was verified and a model of T cell activation stimulated by D antigen was established. Can be used for transfusion immune research.
