以包含BC8241Ht衍生系在内的21个玉米自交系为试材,从DNA分子水平上分析和鉴定欧洲玉米种质BC8241Ht衍生系的遗传多样性与亲缘关系。选取27对SRAP标记引物进行基因组DNA扩增,扩增产生275条谱带,其中有131条带存在多态性,多态性条带比率为47.6%。结果表明:所用21个供试玉米自交系具有丰富的遗传多样性;基于SRAP分子标记的聚类分析将欧洲玉米种质BC8241Ht衍生出的10个玉米自交系聚为一大类,但在聚类树上处于同大类中的第一和第三亚类;欧洲玉米种质BC8241Ht的10个衍生系间存在较近的亲缘关系,但彼此间又表现出一定的遗传差异;在对该种质及其衍生系实施改良过程中,应尽量避免与旅大红骨、塘四平头、PN和西南地方种质混血,可分别用热带或亚热带种质、Lancaster或Reid种质进行改良。
Twenty-one maize inbred lines including BC8241Ht derived lines were used as materials to analyze and identify the genetic diversity and genetic relationship of the European corn germplasm BC8241Ht derived from the molecular level of DNA. Twenty-seven pairs of SRAP-labeled primers were selected for genomic DNA amplification, and 275 bands were amplified. Among them, 131 bands were polymorphic, and the percentage of polymorphic bands was 47.6%. The results showed that the 21 maize inbred lines used in this study had rich genetic diversity. Based on the SRAP molecular marker clustering analysis, 10 maize inbred lines derived from the European corn germplasm BC8241Ht were clustered into one major group. However, The first and the third subclasses in the same taxa of the cluster tree were clustered. There was a close genetic relationship among the 10 derived lines of the European corn germplasm BC8241Ht, but they showed some genetic differences with each other. In the process of improvement of the quality and its derivatives system, we should try to avoid the hybridization with the Luda, Tongsiping, PN and southwest germplasm of Luda, which can be improved respectively with tropical or subtropical germplasm, Lancaster or Reid germplasm.
