在法国,4例用α干扰素(IFN-α)治疗的癌症病人,因心脏病发作而死亡,引起了人们对这种内源性蛋白质毒性的关注。最初,人们推测IFN作为人体自然物质,可能无明显毒性。IFN和白细胞介素(IL)是两类由不同细胞产生和由不同刺激物诱导的自然蛋白质。IFN有潜在的抗病毒和抗增殖特性,并有免疫调节作用。它们在体内的浓度很低,当给予大剂量时,可诱发许多副作用。对肿瘤病人的临床经验表明,所有IFN的副作用和IL-α(不包括液体潴留)的副作用非常相似。
In France, four patients with cancer treated with interferon-alpha (IFN-α) died of a heart attack, causing concern about the endogenous protein toxicity. Initially, it was speculated that IFN, as a natural human substance, may not have significant toxicity. IFN and interleukins (IL) are two natural proteins that are produced by different cells and induced by different stimuli. IFN has potential antiviral and antiproliferative properties and has immunomodulatory effects. Their concentration in the body is very low, and when given in large doses, many side effects can be induced. Clinical experience with cancer patients shows that the side effects of all IFNs and the side effects of IL-α (excluding fluid retention) are very similar.
