脂类是构成细胞内外膜质结构的主要成份。在维持膜结构完整性、信息的跨膜传递和调节代谢等方面起着十分重要的作用。恶性肿瘤时生物膜(包括表面膜、亚细胞膜等)的结构和功能均发生明显改变,导致细胞的各种恶性行为,而这些改变必然伴有脂类组成和代谢的改变。但过去对恶性肿瘤中的蛋白质、酶和核酸研究较多,对脂类则少报道。本实验室曾发现用Solt和Farber法诱发肝癌的鼠肝中,蛋白激酶C在癌前期即见明显升高。因磷脂在蛋白激酶C的调节中起关键作用,故研究肝癌中磷脂的代谢将有助于癌变机理的了解。本文系统地研究了二乙基亚硝胺(DEN)诱发大鼠肝癌过程中,胆固醇和磷脂及其各组分含量的动态变化。
Lipids are the main components that make up the inner and outer membrane structures of cells. It plays a very important role in maintaining the structural integrity of the membrane, transmembrane transmission of information, and regulation of metabolism. In malignant tumors, the structure and function of biofilms (including surface membranes, subcellular membranes, etc.) change significantly, leading to various malignant behaviors of cells, and these changes are necessarily accompanied by changes in lipid composition and metabolism. However, in the past, there have been more studies on proteins, enzymes, and nucleic acids in malignant tumors, and less reports on lipids. In our lab, we found that protein kinase C was significantly increased in the precancerous liver of rat livers induced by Solt and Farber’s method. Since phospholipids play a key role in the regulation of protein kinase C, studying the metabolism of phospholipids in liver cancer will contribute to the understanding of the mechanism of canceration. This article systematically studied the dynamic changes of cholesterol, phospholipids and their components in the process of diethylnitrosamine (DEN)-induced liver cancer in rats.
