目的:探讨喜脑宁冻干粉(XNN)对全脑缺血的保护作用及其可能机制。方法:采用四动脉结扎法建立大鼠全脑缺血/再灌注模型,观察XNN对脑缺血再灌注过程中脑电图及翻正反射恢复时间、脑含水量、脑指数、脑组织匀浆MDA和LD含量、SOD,LDH和NOS活性、海马ET含量的影响;观察XNN对ADP诱导的血小板聚集率的影响;采用Fura-2/AM负载法,观察XNN对血小板内静息[Ca2+]i和CaCl2诱导血小板内[Ca2+]i浓度变化的影响。结果:在全脑缺血再灌注模型上,XNN可以缩短翻正反射恢复时间,促进脑电图的恢复;降低脑含水量、脑指数;提高脑组织SOD,LDH活性,降低LD,MDA含量和NOS活性;降低海马组织ET含量。XNN对ADP诱导的血小板聚集有一定的抑制作用,能降低血小板内静息期[Ca2+]i,并对CaCl2诱导的血小板内[Ca2+]i的升高有抑制作用。结论:XNN对全脑缺血具有一定的保护作用。
Objective: To investigate the protective effect and its possible mechanism of xinaoning lyophilized powder (XNN) on global cerebral ischemia. Methods: The model of global cerebral ischemia / reperfusion was established by four-artery ligation in rats. The changes of electroencephalogram (EEG) and recovery of righting reflex, brain water content, brain index, cerebral homogenate MDA and LD, the activity of SOD, LDH and NOS and the content of ET in the hippocampus were observed. The effect of XNN on ADP-induced platelet aggregation was observed. Fura-2 / AM loading was used to observe the effect of XNN on resting [Ca2 +] i And CaCl2-induced platelet [Ca2 +] i concentration changes. Results: In the model of global cerebral ischemia-reperfusion, XNN can shorten the recovery time of right reflex, promote the recovery of EEG, reduce brain water content and brain index, increase the activities of SOD and LDH, decrease the content of LD and MDA NOS activity; reduce ET content in hippocampus. XNN can inhibit ADP-induced platelet aggregation, decrease platelet resting period [Ca2 +] i, and inhibit CaCl2-induced increase of [Ca2 +] i in platelets. Conclusion: XNN has a protective effect on global cerebral ischemia.
