目的:探讨蜕皮甾酮(Ecdysterone,EDS)对内皮细胞释放一氧化氮的影响。方法:体外培养人脐静脉内皮细胞,测定培养液亚硝酸盐含量。结果:50~400mg/L蜕皮甾酮增加培养上清液中NO2-含量,200mg/L作用达到最高峰;但剂量在800mg/L以上时则减少NO2-含量。EDS对NO2-生成的影响的时效关系为:在2~4h上清液NO2-的含量逐渐增加,在4小时后NO2-含量维持不变。结果:EDS低浓度时促进内皮细胞释放NO,高浓度时抑制内皮细胞释放NO。
Objective: To investigate the effect of Ecdysterone (EDS) on the release of nitric oxide from endothelial cells. METHODS: Human umbilical vein endothelial cells were cultured in vitro and the nitrite content in culture medium was determined. Results: 50-400 mg/L quercetin increased NO2-concentration in the culture supernatant, and the effect of 200 mg/L reached the highest peak; however, when the dose was 800 mg/L or more, the NO2-content was decreased. The time-effect relationship of EDS on the NO2-production was: the NO2-content in the supernatant was gradually increased at 2 to 4 h, and the NO2-content was unchanged at 4 h. RESULTS: EDS promoted the release of NO from endothelial cells at low concentrations, and inhibited the release of NO from endothelial cells at high concentrations.
