纳豆激酶是一新型溶栓药物,它和SubtilisinCarlsberg为同源蛋白.以SubtilisinCarlsberg为模拟蛋白,以AOT/isooctane反胶团体系为有机相,研究了各种萃取条件和反萃取条件对SubtilisinCarlsberg萃取过程的影响.在对模拟体系研究的基础上,利用AOT/isooctane反胶团体系从发酵液中分离纯化纳豆激酶.结果表明,纳豆激酶的萃取行为和SubtilisinCarlsberg非常类似,以目标蛋白的同源蛋白作为模拟蛋白研究萃取规律是合理可行的.纳豆激酶经过一次萃取循环,蛋白质回收率约为33.25%,酶活力回收率达到80.2%,纯化因子约为2.5左右.
Nattokinase is a new type of thrombolytic drug which is homologous to SubtilisinCarlsberg.With SubtilisinCarlsberg as the simulated protein and AOT / isooctane reverse micelle system as the organic phase, the effects of various extraction conditions and back extraction conditions on the SubtilisinCarlsberg extraction process On the basis of the simulation system, the AOT / isooctane reverse micelle system was used to separate and purify nattokinase from the fermentation broth.The results showed that the extraction behavior of nattokinase was very similar to that of SubtilisinCarlsberg, It is reasonable and feasible to study the extraction law of protein as a mimic protein.After an extraction cycle of nattokinase, the recovery rate of protein is about 33.25%, the recovery rate of enzyme activity is 80.2% and the purification factor is about 2.5.
