目的研究缺氧诱导因子-lα(HIF-lα),Caspase-3,Survivin在食管癌的发生、发展中的作用。方法应用免疫组化SP法检测49例食管鳞癌中HIF-lα、Caspase-3、Survivin蛋白的表达。结果HIF-lα蛋白在食管鳞癌组织中的阳性率为51.0%(25/49),显著高于正常食管黏膜0(0/32)(P<0.05);Caspase-3蛋白在食管鳞癌组织中的阳性率为34.7%(17/49),显著低于癌旁正常黏膜100%(32/32)(P<0.05);Survivin蛋白在食管鳞癌组织中的阳性率为57.1%(28/49),显著高于癌旁正常黏膜6.3%(2/32)(P<0.05)。结论HIF-lα蛋白Survivin蛋白的高表达和Caspase-3蛋白的低表达,在食管鳞癌发生、发展中起重要作用,HIF-lα蛋白的阳性表达可能下调下Caspase-3蛋白的表达,使Survivin蛋白的表达增加。联合检测HIF-1α、Caspase-3与Survivin可作为判断食管癌生物学行为的客观指标,对判断食管癌的预后具有重要意义。
Objective To investigate the role of hypoxia inducible factor-1α (HIF-1α), Caspase-3 and Survivin in the pathogenesis and development of esophageal cancer. Methods The expressions of HIF-1α, Caspase-3 and Survivin in 49 cases of esophageal squamous cell carcinoma were detected by immunohistochemical SP method. Results The positive rate of HIF-1α protein in esophageal squamous cell carcinoma was 51.0% (25/49), significantly higher than that in normal esophageal mucosa (0/32) (P <0.05). The positive rate of Caspase-3 protein in esophageal squamous cell carcinoma The positive rate of Survivin in esophageal squamous cell carcinoma was 34.7% (17/49), which was significantly lower than that in the adjacent normal mucosa (32/32) (P <0.05). The positive rate of Survivin protein in esophageal squamous cell carcinoma was 57.1% 49), which was significantly higher than that of normal mucosa (6.3% (2/32)) (P <0.05). Conclusions The high expression of Survivin protein and low expression of Caspase-3 protein in HIF-1α protein play an important role in the occurrence and development of esophageal squamous cell carcinoma. The positive expression of HIF-1α protein may down-regulate the expression of Caspase-3 protein and make Survivin Increased protein expression. The combined detection of HIF-1α, Caspase-3 and Survivin can be used as an objective index to judge the biological behavior of esophageal cancer, which is of great significance in judging the prognosis of esophageal cancer.
