目的:观察脑缺血再灌注大鼠血浆及胞组织内肿瘤坏死因子-α(TNF-α)含量的动态变化。方法:建立大鼠急性不完全性脑缺血再灌注模型.应用放免法(RIA)测定血浆及脑匀浆内TNF-α含量。结果:缺血0.5h,再灌注3h血浆及胞匀浆TNF-α开始明显升高,6h后TNF-α水平达到高峰,随后逐渐下降.但24h后血浆及脑匀浆TNF-α仍维持在较高水平。比较再灌注6h、12h、24h后脑匀浆与血浆TNF-α的水平,发现前者明显高于后者。结论:脑缺血再灌注过程中有TNF-α的升高,且脑组织内TNF-α含量明显高于血浆内水平。
Objective: To observe the dynamic changes of tumor necrosis factor-α (TNF-α) in the plasma and tissue of cerebral ischemia-reperfusion rats. Methods: Acute incomplete cerebral ischemia-reperfusion model was established in rats. The radioimmunoassay (RIA) was used to determine the content of TNF-α in plasma and brain homogenate. Results: The levels of TNF-αin plasma and cell homogenate increased significantly after 0.5h ischemia and 3h reperfusion, and reached the peak at 6h, then decreased gradually. However, the level of TNF-α in plasma and brain homogenate still maintained at a high level after 24h. The levels of TNF-α in brain homogenate and plasma after 6h, 12h, 24h reperfusion were compared, and the former was significantly higher than the latter. CONCLUSION: TNF-α is increased during cerebral ischemia and reperfusion, and the content of TNF-α in brain tissue is significantly higher than that in plasma.
