在改进姐妹染色单体区别染色(SCD)法的研究中,我们继用普通高压水银荧光灯代替国外使用的太阳灯后,制成了高压汞灯。在此基础上,我们对荧光加Giemsa(FPG)法作了较大的改进。即将Hoechst 33258染色与灯照步骤合在一起同步进行。其方法是将Brdu标记好的染色体标本载玻片置于铺锡纸的容器上。滴加4滴Hoechst 33258溶液(用磷酸盐缓冲液配成5μg/ml与25μg/ml溶液),加盖盖玻片。用高压汞灯照射3—20分钟,灯下部距离照射面3.5cm。照毕将盖有盖玻片的一面朝下,放入装有磷酸盐缓冲液的平皿中,
In the study of improving the sister chromatid differential staining (SCD) method, we created a high-pressure mercury lamp following the use of ordinary high-pressure mercury-fluorescent lamps instead of the sun lamps used abroad. On this basis, we have made great improvements to the fluorescence plus Giemsa (FPG) method. Combining Hoechst 33258 dyeing and lighting steps together is synchronized. The method is to place Brdu-labeled slides of chromosomal specimens on containers of tinned paper. Four drops of Hoechst 33258 solution (5 μg/ml with 25 μg/ml solution in phosphate buffer) were added dropwise and the coverslip was capped. The high-pressure mercury lamp is irradiated for 3-20 minutes, and the lower part of the lamp is 3.5 cm away from the irradiated surface.
