建立微波消解-电感耦合等离子体质谱法(ICP-MS),简便、快速、准确地测定大鼠体内铟元素的含量。采用气管灌注法对SPF级雄性Wistar大鼠进行不同浓度的铟锡氧化物染毒。造模结束后处死大鼠,摘取肺脏、肾脏、肝脏、心脏、脾脏、睾丸和股骨。各脏器经微波消解处理后,选用在线内标法及碰撞池/反应池技术校正干扰,采用ICP-MS法测定大鼠体内铟元素的含量。结果显示,该方法能有效消除基体干扰、谱线干扰和物理干扰。测定铟的检出限为0.45 g/L,所检测样品中的最低浓度为0.06 g/L,线性相关系数为0.999 7,精密度为1.09%~3.17%,加标回收率为96.11%~106.43%。提示,ICP-MS法操作简便、准确,可为铟在生物机体内的评价研究提供分析方法参考。
The contents of indium in rats were determined simply, rapidly and accurately by ICP-MS with microwave digestion. SPF male Wistar rats were exposed to different concentrations of indium tin oxide by tracheal perfusion. Rats were sacrificed at the end of modeling and lung, kidney, liver, heart, spleen, testis and femur were harvested. After the organs were treated by microwave digestion, interferences were corrected by online internal standard method and collision cell / reaction cell technique. The contents of indium in rats were determined by ICP-MS. The results show that this method can effectively eliminate the matrix interference, spectral interference and physical interference. The detection limit of indium was 0.45 g / L, the lowest concentration in the sample was 0.06 g / L, the linear correlation coefficient was 0.999 7, the precision was 1.09% ~ 3.17%, the recovery was 96.11% ~ 106.43 %. Tip, ICP-MS method is simple and accurate, and can provide an analytical method for the evaluation of indium in vivo.
