清瘟败毒饮影响急性肺损伤大鼠肺组织NF-κBp65表达的实验研究

目的:通过观察清瘟败毒饮对脂多糖(LPS)诱导急性肺损伤(ALI)大鼠肺组织病理及核因子-κBp65(NF-κBp65)表达的影响,探讨清瘟败毒饮对ALI的治疗作用及干预机制。方法:选取健康雄性SD大鼠144只,随机分为6组,每组24只,分别为正常对照组,模型组,泼尼松组和清瘟败毒饮大、中、小剂量组。脂多糖溶液(2mg/kg)气管内滴注复制ALI大鼠模型,连续两天。各治疗组在第2次滴注完LPS溶液1h后,用生理盐水将相应的药物稀释至4mL灌胃,1天2次。各组分别于灌胃后24h、48h处死6只大鼠,于72h处死余下所有大鼠,并收集所需标本;通过镜下观察肺组织病理变化及采用W estern b lot方法检测肺组织中NF-κBp65的表达水平。结果:LPS致ALI大鼠肺组织肺泡结构破坏或实变,肺间质明显水肿,大量炎性细胞浸润,肺泡腔和肺间质内可见局灶性炎症细胞浸润和大量红细胞渗出,与同时相模型组比较,清瘟败毒饮大、中剂量组大鼠病理学炎症积分均降低非常显著(P<0.01),而小剂量组只有24h、48h有显著性降低或非常显著性降低(P<0.05或P<0.01);清瘟败毒饮各剂量治疗组中48h清瘟败毒饮大、中剂量组和72h各剂量组大鼠肺组织中NF-κBp65的表达均降低,与同时相模型组相比均有显著性差异或非常显著性差异(P<0.05或P<0.01)。结论:清瘟败毒饮能减轻LPS致ALI大鼠肺组织的损伤程度,在一定程度上减少炎症细胞在肺部积聚、浸润、渗出,起到比较明确的肺保护作用;清瘟败毒饮能有效减少LPS致ALI大鼠肺组织中NF-κBp65的表达,通过抑制NF-κB的活化和炎性细胞因子的生成,对炎症反应起到抑制作用;在LPS致ALI早期使用清瘟败毒饮,可有效减轻肺组织的损伤程度,减轻肺部的炎症反应。 OBJECTIVE: To observe the effect of Qingyan Baidu decoction on the lung histopathology and the expression of nuclear factor-κBp65 in LPS-induced acute lung injury (ALI) rats and to investigate the effect of Qingwen Baidu decoction on ALI Therapeutic effect and intervention mechanism. Methods: A total of 144 healthy male Sprague-Dawley rats were randomly divided into 6 groups (24 rats in each group), which were normal control group, model group, prednisone group, and Qingwenbaidundan large, medium and small dose groups respectively. Lipopolysaccharide solution (2mg / kg) tracheal instillation copy ALI rat model for two consecutive days. Each treatment group after the second instillation of LPS solution 1h, the corresponding drug diluted with normal saline to 4mL intragastric administration, 2 times a day. Six rats in each group were sacrificed at 24h and 48h after gavage respectively. All the rats were sacrificed at 72h, and the required specimens were collected. Pathological changes of lung tissue were observed by microscopy and the expression of NF -κBp65 expression levels. RESULTS: The alveolar structure was destroyed or solidified in the lung tissue of ALI rats induced by LPS, the interstitial edema, a large number of inflammatory cell infiltration, infiltration of inflammatory cells and a large number of erythrocytes were seen in alveolar space and pulmonary interstitium, Compared with the phase model group, the pathological inflammation scores of the rats in the middle-dose group and the middle-dose group of Qingwen Baidu Decoction decreased significantly (P <0.01), while those in the low-dose group decreased significantly or significantly (P <0.05 or P <0.01). The expression of NF-κBp65 in lung tissue of rats in each dose group of Qingwen Baidu Decoction group for 48h was significantly lower than that of the control group There was significant difference or very significant difference between model group (P <0.05 or P <0.01). Conclusion: Qingwen Baidu Decoction can reduce the degree of lung injury induced by LPS in ALI rats, and reduce the accumulation, infiltration and exudation of inflammatory cells to a certain extent, and play a more specific role in lung protection. Drink can effectively reduce the LPS-induced ALI rat lung tissue NF-κBp65 expression, by inhibiting the activation of NF-κB and inflammatory cytokine production, play an inhibitory effect on inflammatory response; Toxic drinking can effectively reduce the degree of lung tissue damage and reduce lung inflammation.
