目的观察1,25-二羟维生素D3对体外培养的人成骨样细胞增殖与分化的影响,以进一步探讨其治疗骨质疏松的机制。方法采用MTT比色分析法检测1,25-二羟维生素D3对体外培养OS—732人成骨样细胞增殖的作用与效价。采用培养细胞匀浆上清测单位蛋白下的碱性磷酸酶活性,培养基中骨钙素(BGP)含量分析方法检测1,25-二羟维生素D3对OS—732人成骨样细胞分化的影响。结果培养第4天,10-7mol/L1,25-二羟维生素D3显著抑制成骨样细胞增殖(P<0.05),10-8mol/L、10-9mol/L1,25-二羟维生素D3对成骨样细胞的作用不明显。培养4天后培养基中BGP水平在10-7mol/L组变化不大,在10-8mol、10-9mol/L1,25-二羟维生素D3组BGP水平显著增高(P<0.05,P<0.01)。结论1,25-二羟维生素D3抑制成骨样细胞增殖在高剂量时作用显著,促进成骨样细胞分化作用在较低剂量显著。
Objective To investigate the effects of 1,25-dihydroxyvitamin D3 on the proliferation and differentiation of human osteoblast-like cells in vitro in order to further explore the mechanism of osteoporosis treatment. Methods MTT colorimetric assay was used to detect the effect of 1,25-dihydroxyvitamin D3 on proliferation of OS-732 human osteoblast-like cells in vitro. Alkaline phosphatase activity was measured using the supernatant of cultured cell homogenate and the osteocalcin (BGP) content in culture medium was assayed for differentiation of OS-732 human osteoblast-like cells by 1,25-dihydroxyvitamin D3 influences. Results On the fourth day after culture, 10-7mol / L, 25-dihydroxyvitamin D3 significantly inhibited osteoblast-like cell proliferation (P <0.05), 10-8mol / L, 10-9mol / L, 25-dihydroxyvitamin D3 Osteoblast-like cells have no obvious effect. After 4 days of culture, the level of BGP in the culture medium did not change much in 10-7mol / L group, and the BGP level in 10-8mol, 10-9mol / L1, 25-dihydroxyvitamin D3 group was significantly increased (P <0.05, P <0.01) . Conclusion 1,25-dihydroxyvitamin D3 can inhibit the proliferation of osteoblast-like cells at high doses and promote the differentiation of osteoblast-like cells at lower doses.
