目的试图从霉菌培养液中分离纯化抗真菌活性蛋白质。方法用离子交换层析法分离,用反 向高效液相色谱法进一步纯化,经N-三(羟甲基)甲基甘氨酸凝胶电泳分析确定其相对分子质量。结果从黑曲霉 菌培养上清液中分离出一种具有抗真菌活性的蛋白质(AFP)。纯化后得到高活性的精制AFP。相对分子质量约为 9000,经MTT法检测5种真菌具有明显抗真菌活性。结论本研究为抗菌药物及防腐剂的开发提供了依据。
Objective To isolate and purify antifungal active protein from mold culture broth. The method was separated by ion exchange chromatography and further purified by reverse-phase high performance liquid chromatography. The relative molecular mass was determined by N-tris (hydroxymethyl) methylglycine gel electrophoresis. As a result, an antifungal protein (AFP) was isolated from Aspergillus niger culture supernatant. After purification, highly active purified AFP was obtained. The relative molecular mass of about 9000, by MTT assay of five fungi have significant antifungal activity. Conclusion This study provides the basis for the development of antimicrobial agents and preservatives.
