目的 探讨中国 2型糖尿病患者付氧酶 (PON)基因与并发冠心病 (CAD)的关系。方法利用聚合酶链反应—变性梯度凝胶电泳技术对 49例 2型糖尿病合并CAD患者、49例未合并CAD的2型糖尿病对照者和 10 1例健康对照者进行PON基因外显子筛查。结果 发现中国人群PON基因第 191位密码子存在Gln191 Arg多态性 ,等位基因以A/B表示。CAD患者PON基因的 3种基因型(AA、AB和BB)的构成比与 2型糖尿病对照和健康对照差异有显著性 ,B等位基因频率较两对照组显著增高 (0 .79比 0 .6 2和 0 .6 1,均P <0 .0 1)。结论 中国 2型糖尿病患者PON基因第 191位密码子的Gln191 Arg多态性与并发CAD有关 ,B等位基因是 2型糖尿病并发CAD的危险因素之一。
Objective To investigate the relationship between PON gene and CAD in Chinese patients with type 2 diabetes mellitus (T2DM). Methods 49 cases of type 2 diabetes mellitus complicated with CAD, 49 cases of type 2 diabetes without CAD and 101 healthy controls were enrolled for exon screening using polymerase chain reaction - denaturing gradient gel electrophoresis. The results showed that Gln191 Arg polymorphism was found in codon 191 of Chinese PON gene and the allele was A / B. The proportions of three genotypes (AA, AB and BB) of PON gene in CAD patients were significantly different from those in T2DM controls and healthy controls. The frequency of allele B in CAD patients was significantly higher than that in controls (0.79 vs 0.79). 6 2 and 0 .6 1, both P <0. 0 1). Conclusion Gln191 Arg polymorphism of codon 191 in PON gene is associated with complicated CAD in type 2 diabetes mellitus in China. B allele is one of the risk factors of type 2 diabetes with CAD.
