目的 用 5 溴 2’脱氧脲嘧啶核苷 (BrdU)标记法观察大鼠胰腺导管中成体干细胞的增殖变化情况 ,探索其变化规律。方法 自鼠尾静脉注入链脲菌素 (STZ)造成大鼠胰岛 β细胞损伤后 ,于注射后第 1、2、3、6、9天分别取 3只大鼠处死 ,处死前 12h ,腹腔注入 5 溴 2’脱氧脲嘧啶核苷 (BrdU)标记胰腺导管上皮细胞中s期细胞 ,用免疫组织化学法观察胰腺导管上皮中干细胞的增殖变化。结果 注入链脲菌素后 ,大鼠的导管上皮细胞显著增殖 (P <0 .0 0 1) ,于 48h迅速达到高峰 ,分类导管中主导管、中导管、小导管于 48h达到高峰 ,共同导管于 72h达到高峰。结论 胰岛 β细胞的损伤可以激活胰腺导管中的干细胞 ,于损伤后 48h细胞增殖最为活跃。
OBJECTIVE: To observe the proliferation and proliferation of adult rat pancreatic duct by BrdU labeling method and explore its variation. Methods After the islet β cell injury was induced by injection of streptozotocin (STZ) into the tail vein of rats, 3 rats were sacrificed at 1, 2, 3, 6, and 9 days after injection. S-phase cells were stained with 5-bromo-2’-deoxyuridine (BrdU) in pancreatic ductal epithelial cells and the proliferation of pancreatic ductal epithelium was observed by immunohistochemistry. Results After administration of streptozotocin, the ductal epithelial cells of rats significantly proliferated (P <0.01), reaching a peak at 48 hours. The main duct, middle duct, and small duct of the classified duct reached a peak at 48 hours. The common duct Peak at 72h. Conclusion The islet β cell injury can activate the stem cells in the pancreatic duct, and the cell proliferation is the most active 48 h after injury.
