目的以内皮素(ET)及一氧化氮(NO)为心室重构观察指标研究贝那普利联合螺内酯应用对心室重构大鼠的影响。方法结扎Wistar雄性大鼠腹主动脉建立压力超负荷性心室重构大鼠模型。48只建模后存活大鼠随机分为4组(每组12只):贝那普利联合螺内酯组(贝那普利10mg·kg-1.d-1,螺内酯15mg·kg-1.d-1),贝那普利组(贝那普利20mg·kg-1.d-1),螺内酯组(螺内酯30mg·kg-1.d-1),生理盐水组(生理盐水10ml.kg-1.d-1);不结扎腹主动脉的12只作为假手术组(蒸馏水10ml.kg-1.d-1);各组每天灌胃给药1次,共6周。给药6周后用放免法测定心肌中ET和血浆中NO含量。结果建模组与假手术组相比,心肌中ET含量明显升高(P<0.05),血浆NO水平明显降低(P<0.05),表明大鼠心室重构模型建立成功。贝那普利联合螺内酯组及贝那普利、螺内酯单独给药组均能降低心肌ET的含量及升高血浆中NO含量(P<0.05或P<0.01),但贝那普利联合螺内酯组比贝那普利组、螺内酯组作用更明显(P<0.05)。结论联合应用贝那普利与螺内酯可更有效逆转大鼠心室重构。
Objective To investigate the effects of benazepril and spironolactone on ventricular remodeling in rats with ventricular remodeling using endothelin (ET) and nitric oxide (NO) as ventricular remodeling indicators. Methods A rat model of pressure overload was established by ligating the abdominal aorta of Wistar male rats. Forty-eight survived rats were randomly divided into 4 groups (12 in each group): benazepril combined with spironolactone group (benazepril 10mg · kg-1.d-1, spironolactone 15mg · kg-1.d -1), benazepril group (benazepril 20 mg · kg -1 · d -1), spironolactone group (spironolactone 30 mg · kg -1 · d -1), saline group (10ml · kg- 1.d-1); 12 rats without ligating abdominal aorta as sham operation group (distilled water 10ml.kg-1.d-1); each group was given gavage once a day for 6 weeks. After 6 weeks of administration, the levels of ET and plasma NO in myocardium were determined by radioimmunoassay. Results Compared with the sham operation group, the content of ET in myocardium was significantly increased (P <0.05) and the level of NO in plasma was significantly decreased (P <0.05), which indicated that the rat ventricular remodeling model was established successfully. The combination of benazepril and spironolactone and benazepril and spironolactone alone could decrease ET content and increase NO content in plasma (P <0.05 or P <0.01), but benazepril combined with spironolactone Compared with benazepril group and spironolactone group, the effect was more obvious (P <0.05). Conclusions Combination of benazepril and spironolactone can effectively reverse ventricular remodeling in rats.
