酯酶染色法作为T淋巴细胞的一种新的鉴别技术有一定实用价值。本文着重介绍了将该方法用于实验小鼠,并在过去实验的基础上重新探讨并对染色方法做了改进,即:1)固定液必须新鲜,2)甲基绿要用氯仿提纯,3)血涂片孵育需适宜的温度和湿度。改进后的方法染色效果满意,镜下小鼠末稍血T淋巴细胞清晰。为证实本法染色细胞系T细胞,设计了用X线照射小鼠胸腺区的试验,然后再检查T淋巴细胞,结果表明本染色法是可靠的。该法最大优点是取血量少,且能够做多次连续性实验观察,在染色方法上比较简便,易于掌握,一般实验室均可做到。
Esterase staining as T lymphocytes a new identification technology has some practical value. The improved method of staining was satisfactory, microscopic T lymphocytes in mice after a clear end. In order to confirm the staining of cell line T cells by this method, we designed the experiment of irradiating the thymus area of mice with X-rays and then examining the T lymphocytes. The result shows that this staining method is reliable. The biggest advantage of this method is less blood, and can do a number of continuous experimental observation, the staining method is relatively simple and easy to grasp, the general laboratory can be done.
