目的探讨线粒体生物合成相关基因PGC-1α、PGC-1β、NRF1、NRF2和ERRαmRNA在子痫前期(PE)患者胎盘组织中的表达与意义。方法采用RT-PCR技术测定36例正常足月妊娠妇女(A组)和24例轻度PE患者(B组)及17例重度PE患者(C组)胎盘组织中PGC-1α、PGC-1β、NRF1、NRF2和ERRαmRNA表达水平。结果 C组PGC-1α、PGC-1β、NRF1、NRF2和ERRαmRNA表达水平分别为0.43±0.13、0.20±0.09、0.29±0.13、0.71±0.14和0.48±0.13,B组分别为0.78±0.13、0.37±0.10、0.96±0.19、0.81±0.22和1.08±0.16,均显著低于A组的1.47±0.19、0.80±0.14、1.84±0.22、1.59±0.18和1.54±0.21(P<0.05)。C组PGC-1α、PGC-1β、NRF1和ERRαmRNA表达水平显著低于B组(P<0.05)。结论上述线粒体生物合成相关基因在PE患者胎盘组织中的差异表达参与了PE的病理生理过程。
Objective To investigate the expression and significance of mitochondrial PGC-1α, PGC-1β, NRF1, NRF2 and ERRαmRNA in placenta of patients with preeclampsia (PE). Methods The levels of PGC-1α, PGC-1β, P-selectin in placenta from 36 normal pregnant women (group A), 24 mild PE patients (group B) and 17 severe PE patients (group C) NRF1, NRF2 and ERRα mRNA expression levels. Results The expressions of PGC-1α, PGC-1β, NRF1, NRF2 and ERRαmRNA in group C were 0.43 ± 0.13,0.20 ± 0.09,0.29 ± 0.13,0.71 ± 0.14 and 0.48 ± 0.13, respectively, while those in group B were 0.78 ± 0.13 and 0.37 ± 0.13 0.10, 0.96 ± 0.19, 0.81 ± 0.22 and 1.08 ± 0.16, all significantly lower than 1.47 ± 0.19, 0.80 ± 0.14, 1.84 ± 0.22, 1.59 ± 0.18 and 1.54 ± 0.21 in group A (P <0.05). The expression of PGC-1α, PGC-1β, NRF1 and ERRαmRNA in group C was significantly lower than that in group B (P <0.05). Conclusion The differential expression of mitochondrial biosynthesis genes in the placenta of PE patients is involved in the pathophysiological process of PE.
