为了提高小麦幼胚体细胞再生频率,以河南省大面积推广的豫麦49号、豫麦18号和兰考906为研究对象,研究了不同浓度AgNO3对小麦幼胚胚性愈伤组织培养特性和草酸盐氧化酶活性的影响。结果表明:基因型间、不同AgNO3浓度间小麦幼胚胚性愈伤组织诱导率、愈伤组织绿点率和草酸盐氧化酶活性差异均达到0.01显著水平,基因型间以豫麦18最高、兰考906最低;不同AgNO3浓度间以MS培养基上附加2.5~5.0 mg/L处理较高。不同浓度Ag-NO3条件下小麦幼胚胚性愈伤组织诱导率与草酸盐氧化酶活性相关系数为0.920,达0.01显著水平。可见,在小麦幼胚组织培养过程中可以加入Ag+提高再生频率。
In order to improve the frequency of somatic embryogenesis in wheat immature embryos, we studied the effects of different concentrations of AgNO3 on the embryogenic callus traits of wheat immature embryos and the effects of different concentrations of AgNO3 on Yumai 49, Yumai 18 and Lankao 906, Oxalate oxidase activity. The results showed that the differences of embryogenic callus induction, green spot callus rate and oxalate oxidase activity in wheat immature embryos between genotypes with different concentrations of AgNO3 reached 0.01 significant level, and the highest among Yumai 18 , Lankao 906 lowest; different AgNO3 concentration to MS medium supplemented with 2.5 ~ 5.0 mg / L treatment is higher. The correlation coefficient of oxalic acid oxidase activity and the induction rate of embryogenic callus of wheat immature embryos under different concentrations of Ag-NO3 was 0.920, reaching a significant level of 0.01.
