目的 利用全反式维甲酸 (ATRA)诱导Wistar大鼠 ,建立马蹄内翻足动物模型 ,分析ATRA最适诱导剂量。方法 将Wistar孕鼠随机分为 4个用药组及对照组 ,孕 10d分别按 12 0 ,130 ,135 ,14 0mg/(kg·bw)不同剂量经口给予ATRA矿物油混悬液和纯矿物油。孕 2 1d剖检胎鼠 ,并对足踝部切片观察。结果 ATRA可诱导Wistar大鼠胚胎发生马蹄内翻足样畸形及其它畸形 ,发生率随剂量的增加而增高 ;单纯马蹄内翻足样畸形的发生率 135mg/(kg·bw)组为最高 ,达 2 9 2 7%。结论 135mg/(kg·bw)ATRA是诱导马蹄内翻足样畸形的理想剂量。
Objective To establish an animal model of clubfoot by using all-trans retinoic acid (ATRA) -induced Wistar rats and analyze the optimal induction dose of ATRA. Methods Pregnant Wistar rats were randomly divided into four groups: control group and control group. The pregnant rats were orally administered ATRA mineral oil suspension and pure mineral oil at different doses of 12 0, 130, 135 and 140 mg / (kg · bw) . Pregnancy 21d dissection of fetal rats, and the ankle section observation. Results ATRA could induce foot-like deformity and other deformities in Wistar rats. The incidence rate of ATRA increased with the increase of dose. The incidence of pure clubfoot 135mg / (kg · bw) was the highest 2 9 2 7%. Conclusion 135 mg / (kg · bw) ATRA is the ideal dose for inducing clubfoot deformity.
