目的:建立自体髓核移植致大鼠非压迫腰椎间盘髓核突出神经根性疼痛模型,探讨优化活血利水方对移植髓核MMP-3、TIMP-1表达的影响。方法:40只SD大鼠随机分为对照组、模型组、中药组、阿斯匹林组,建立大鼠自体髓核移植致腰5神经根性痛模型,造模后1周起各组分别予生理盐水1.82mL、生理盐水1.82mL、中药汤剂1.82mL、阿斯匹林1.82mL(0.1g.kg-1.d-1),每日1次,连续用药4周。第31天取移植髓核检测MMP-3、TIMP-1mRNA及蛋白的表达。结果:正常髓核中MMP-3、TIMP-1mRNA及蛋白表达量很少,各实验组移植髓核中MMP-3、TIMP-1mRNA及蛋白表达量与对照组比较均显著增多(P<0.0l)。与模型组比较,中药组、西药组MMP-3mRNA及蛋白表达量显著减少(P<0.01),TIMP-1mRNA及蛋白相对表达量显著增多(P<0.01)。中、西药组比较,两组间存在显著性差异,中药组MMP-3mRNA及蛋白显著减少,TIMP-1mRNA及蛋白显著增多。结论:优化活血利水方可以显著下调移植髓核MMP-3mRNA及其蛋白表达,上调TIMP-1mRNA及其蛋白表达,减轻椎间盘的退变,促进椎间盘的修复。
OBJECTIVE: To establish a model of radicular nerve root pain induced by autologous nucleus pulposus transplantation in non-compressed lumbar disc nucleus in rats and to investigate the effect of Huoxuoli Recipe on the expression of MMP-3 and TIMP-1 in nucleus pulposus. Methods: Forty SD rats were randomly divided into control group, model group, TCM group and aspirin group. The model of lumbar 5 nerve root pain induced by autologous nucleus pulposus was established in rats. One week after modeling, 1.82 mL of physiological saline, 1.82 mL of physiological saline, 1.82 mL of traditional Chinese medicine decoction, and 1.82 mL of aspirin (0.1 g.kg-1.d-1) once daily for 4 weeks. On the 31st day, the nucleus accumbens were taken to detect the expression of MMP-3 and TIMP-1 mRNA and protein. Results: The mRNA and protein expressions of MMP-3 and TIMP-1 in the normal nucleus pulposus were very low. The mRNA and protein expressions of MMP-3 and TIMP-1 in the nucleus pulposus of each experimental group were significantly increased compared with the control group (P <0.01 ). Compared with model group, the expression of MMP-3mRNA and protein in Chinese medicine group and western medicine group were significantly decreased (P <0.01), and the relative expression of TIMP-1 mRNA and protein was significantly increased (P <0.01). There was a significant difference between the two groups in the western medicine group and the MMP-3 mRNA and protein in the Chinese medicine group were significantly reduced, TIMP-1 mRNA and protein were significantly increased. CONCLUSION: Optimizing Huoxue Recipe can significantly down-regulate the expression of MMP-3mRNA and its protein in the nucleus pulposus, increase the expression of TIMP-1mRNA and its protein, relieve the degeneration of intervertebral disc and promote the repair of intervertebral disc.
