目的:探讨原花青素(PC)预处理对体外人淋巴母细胞AHH-1和肠腺上皮细胞HIEC电离辐射损伤的防护效果及可能的作用机制。方法:采用CCK-8法检测原花青素预处理对γ射线照射后AHH-1、HIEC细胞存活率的影响,Annexin-Ⅴ/PI染色和流式细胞术分析细胞凋亡及细胞周期的变化,蛋白质印迹法检测细胞Bcl-2蛋白表达变化。结果:PC预处理的细胞辐照后存活率明显升高(P<0.01),4Gy照射用药组AHH-1细胞凋亡率(11.78%)显著低于单纯照射组(26.38%,P<0.01),8Gy照射用药组HIEC细胞凋亡率(5.32%)亦显著低于单纯照射组(12.45%,P<0.01)。PC预处理组AHH-1、HIEC细胞受照射后其Bcl-2蛋白表达下降受到抑制。结论:PC预处理对细胞的辐射损伤具有较好的防护效果,可能与其促进受损细胞Bcl-2蛋白表达有关。
Objective: To investigate the protective effect and possible mechanism of procyanidins (PC) pretreatment on human lymphoblasts AHH-1 and intestinal epithelial cells HIEC in vitro. METHODS: CCK-8 method was used to detect the effect of procyanidins pretreatment on the survival rate of AHH-1 and HIEC cells after γ-irradiation. Annexin-V/PI staining and flow cytometry were used to analyze apoptosis and cell cycle changes. Western blot The method was used to detect the changes of Bcl-2 protein expression. RESULTS: The survival rate of PC-pretreated cells was significantly increased after irradiation (P<0.01), and the apoptosis rate of AHH-1 cells (11.78%) was significantly lower in the 4 Gy irradiation group than in the simple irradiation group (26.38%, P<0.01). The rate of apoptosis of HIEC cells in the 8Gy irradiation group (5.32%) was also significantly lower than that in the group irradiated (12.45%, P<0.01). In the PC pretreatment group, the decrease of Bcl-2 protein expression was inhibited after irradiation of AHH-1 and HIEC cells. Conclusion: PC pretreatment has a good protective effect on cell radiation damage, which may be related to the promotion of Bcl-2 protein expression in damaged cells.
