目的:研究并比较DREAM蛋白的2个主要亚型DREAM A和DREAM B在小鼠多种组织尤其是神经组织的表达水平的差异。方法:取不同年龄小鼠的各种组织匀浆后提取RNA,采用real time PCR的方法检测各种组织中DREAM蛋白2个亚型的mRNA的表达水平。结果:DREAM的两个亚型A和B在神经和非神经组织中广泛存在。在大脑皮层、小脑、海马及下丘等神经组织中DREAM A的表达水平要远高于DREAM B,并且在小鼠上述神经组织中DREAM A随着动物发育成熟,表达水平会逐渐升高,而DREAM B在神经组织发育初期呈较高水平表达。结论:DREAM A是成年小鼠中DREAM的主要亚型,而DREAM B可能在动物的发育早期发挥一定作用。
OBJECTIVE: To study and compare the differences in the expression levels of DREAM A and DREAM B, two major subtypes of DREAM protein in various tissues of mice, especially in neural tissues. Methods: RNA was extracted from various tissue homogenates of different age mice, and mRNA expression levels of two subtypes of DREAM protein in various tissues were detected by real time PCR. RESULTS: The two subtypes A and B of DREAM are widespread in both neuronal and non-neural tissues. The expression level of DREAM A in nerve tissue such as cerebral cortex, cerebellum, hippocampus and inferior colliculus is much higher than that of DREAM B, and the expression level of DREAM A gradually increases as the animal matures in the above nerve tissues of mice DREAM B showed a high level of expression in the early stage of nerve tissue development. CONCLUSIONS: DREAM A is a major subtype of DREAM in adult mice, whereas DREAM B may play a role early in animal development.
