目的观察环氧合酶-2(COX-2)选择性抑制剂NS-398对食管癌细胞株EC9706增殖及凋亡的影响,研究其对凋亡抑制蛋白Survivin和Caspase-3表达的影响,探讨NS-398诱导EC9706细胞凋亡的作用机制。方法NS-398作用EC9706细胞后,MTT法测定NS-398对人食管癌EC9706细胞增殖的抑制率;DNA片段分析法和流式细胞仪检测细胞凋亡;免疫细胞化学检测Survivin和Caspase-3蛋白表达变化。结果NS-398(10~100μmol/L)对EC9706细胞生长有抑制作用,随浓度升高、时间延长抑制作用增强,并诱导EC9706细胞凋亡,呈剂量-时间效应关系;NS-398可降低Survivin蛋白表达,增加Caspase-3蛋白表达。结论NS-398可诱导人食管癌细胞株EC9706凋亡,其机制可能与下调Survivin表达及激活Capase-3表达有关。
Objective To investigate the effects of cyclooxygenase-2 (COX-2) selective inhibitor NS-398 on the proliferation and apoptosis of esophageal carcinoma cell line EC9706 and to investigate its effect on the expression of Survivin and Caspase-3 Mechanism of NS-398 inducing apoptosis in EC9706 cells. Methods The inhibitory effect of NS-398 on the proliferation of human EC9706 cells was determined by MTT assay after NS-398 cells were treated with NS-398. The apoptosis of EC9706 cells was detected by DNA fragmentation assay and flow cytometry. The expressions of Survivin and Caspase-3 protein Change of expression Results NS-398 (10-100μmol / L) inhibited the growth of EC9706 cells. With the increase of concentration and time, NS-398 increased the inhibitory effect and induced the apoptosis of EC9706 cells in a time-dose-dependent manner. NS-398 decreased the expression of Survivin Protein expression, increase Caspase-3 protein expression. Conclusion NS-398 can induce apoptosis of human esophageal cancer cell line EC9706, which may be related to the down-regulation of Survivin expression and the activation of Capase-3 expression.
