本研究采用碱性彗星试验评价个体外周血淋巴细胞DNA损伤,聚合酶链反应-限制性片段长度多态性方法分析细胞色素P450 2E1(CYP2E1)、微粒体环氧化物水解酶(m EH)、谷胱甘肽S-转移酶P1(GSTP1)基因的多态性,探讨外源性化学物代谢酶基因多态性与1,3-丁二烯(BD)工人外周血淋巴细胞DNA损伤的关系。结果发现,BD暴露组和对照组彗星尾距分别为4.65(3.49~5.98)和2.38(0.82~3.98),差异具有统计学意义(P<0.01)。暴露组中具有CYP2E1基因Pst1位点c1/c2或c2/c2基因型个体的彗星尾距显著高于c1/c1基因型个体(P<0.01)。
In this study, alkaline comet assay was used to evaluate the DNA damage of peripheral blood lymphocytes and the expression of cytochrome P450 2E1 (CYP2E1) and microsomal epoxide hydrolase (mEH) were analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) Glutathione S-transferase P1 (GSTP1) gene polymorphism and explore the exogenous chemical metabolizing enzyme gene polymorphism and 1,3-butadiene (BD) workers with peripheral blood lymphocyte DNA damage . The results showed that the comet tail distances of BD exposure group and control group were 4.65 (3.49 ~ 5.98) and 2.38 (0.82 ~ 3.98), respectively, with statistical significance (P <0.01). The comet tail distance of c1 / c2 or c2 / c2 genotypes with Pst1 locus of CYP2E1 gene in the exposed group was significantly higher than that in c1 / c1 genotype individuals (P <0.01).
