目的:检测Notch受体在肝癌组织中的表达水平,为肝癌的治疗提供新的靶点。方法:应用免疫组化和荧光定量PCR(FQ-PCR)技术对35例肝癌组织和13例正常肝组织中的Notch1、Notch2和Notch4受体蛋白及基因表达情况进行检测分析。结果:免疫组织化学结果表明,Notch分子在肝癌组织中存在强弱不等的表达,Notch1和Notch4表达上调,Notch2表达下调,差异有统计学意义,P<0.05;FQ-PCR技术检测显示,肝癌患者Notch1和Notch4基因表达阳性,DNA拷贝数显著高于对照组,差异有统计学意义,P<0.05;肝癌组Notch2基因表达阴性,显著低于正常对照组(P<0.05);Notch分子的蛋白表达水平与基因表达的拷贝数呈正相关,r=0.985,0.664。结论:Notch受体与肝癌的发生、发展密切相关,Notch受体在肝癌发生、发展中的作用机制将为肝癌的基因治疗及新药开发提供理论依据。
Objective: To detect the expression of Notch receptor in hepatocellular carcinoma and to provide a new target for the treatment of liver cancer. Methods: The protein and gene expression of Notch1, Notch2 and Notch4 in 35 cases of HCC and 13 cases of normal liver tissues were detected by immunohistochemistry and fluorescence quantitative PCR (FQ-PCR). Results: The results of immunohistochemistry showed that Notch molecules expressed in varying degrees of strength in HCC tissues. Notch1 and Notch4 expressions were up-regulated and Notch2 expression was down-regulated. There was significant difference between them (P <0.05). FQ-PCR showed that HCC Notch1 and Notch4 gene expression was positive in patients with DNA copy number was significantly higher than the control group, the difference was statistically significant, P <0.05; Notch2 gene expression was negative in hepatocellular carcinoma was significantly lower than the normal control group (P <0.05); Notch protein The expression level was positively correlated with the copy number of gene expression, r = 0.985,0.664. Conclusion: Notch receptor is closely related to the occurrence and development of hepatocellular carcinoma. The mechanism of action of Notch receptor in the occurrence and development of hepatocellular carcinoma will provide a theoretical basis for the gene therapy of hepatocellular carcinoma and the development of new drugs.
