目的:从形态学角度观察探讨自由基清除剂对PrP106-126诱导的分化PC12细胞损伤的保护作用。方法:应用光镜、激光共聚焦显微镜技术和MTT法,体外观察不同浓度自由基清除剂对PrP106-126诱导的分化PC12细胞形态学及细胞存活率改变的影响。结果:不同浓度的依达拉奉(15、30、45、60μmol·L-1)对诱导分化的PC12细胞均有保护作用,其保护作用呈剂量依赖关系。激光共聚焦显微镜双荧光标记结果显示,随着依达拉奉浓度的增高,PrP106-126诱导的分化PC12细胞坏死及凋亡的程度降低。结论:依达拉奉对PrP106-126诱导的分化PC12细胞具有抗氧化损伤保护作用。
OBJECTIVE: To observe the protective effect of free radical scavengers on the differentiation of PC12 cells induced by PrP106-126 from the morphological point of view. Methods: The morphological and cell viability of PC12 cells induced by PrP106-126 were observed in vitro by light microscopy, laser confocal microscopy and MTT assay. Results: Edaravone (15,30,45 and 60μmol·L-1) had protective effects on the differentiation of PC12 cells in a dose-dependent manner. Double-labeling by laser scanning confocal microscopy showed that Prd106-126 induced a decrease in the degree of necrosis and apoptosis of differentiated PC12 cells with the increase of edaravone concentration. Conclusion: Edaravone can protect PC12 cells induced by PrP106-126 against oxidative damage.
