目的从免疫学角度分析五味子多糖抑制肝癌小鼠肿瘤生长的作用机理。方法选取健康小鼠60只,随机分为5组,各组12只。对照组常规饲养,日灌予等剂量的生理盐水;其余4组以H22肝癌细胞株接种,制备肝癌模型,模型组日给予等剂量的生理盐水;高、中、低剂量五味子多糖组分别灌予40 mg·kg-1、20 mg·kg-1、10 mg·kg-1的五味子多糖;1周后取下腔静脉血,分离血清,以流式细胞仪测定免疫细胞水平;同时,剥离瘤体、脾脏和胸腺,计算抑瘤率、脾脏指数和胸腺指数。结果与对照组相比,模型组小鼠的血清免疫细胞水平及胸腺指数均有显著降低(P<0.05),脾脏指数无明显差异(P>0.05);与模型组相比,五味子低、中、高剂量小鼠的血清免疫细胞水平及胸腺指数均有显著升高(P<0.05或P<0.01),且升高幅度随着给药剂量的增加而增大,其中高剂量组的血清免疫细胞水平及胸腺指数与对照组无显著差异(P>0.05)。经计算,五味子多糖低、中、高剂量组的抑瘤率分别为18.7%、41.1%、61.4%。结论五味子多糖可同时提高小鼠细胞免疫及器官免疫功能,从而起到一定的肿瘤抑制效应,且该效应在一定范围内呈现出典型的剂量依赖性,随着给药剂量的增加而增强。
Objective To analyze the mechanism of Schisandra chinensis polysaccharides inhibiting the tumor growth of hepatocellular carcinoma mice from immunological point of view. Methods Sixty healthy mice were randomly divided into 5 groups, 12 in each group. The rats in the control group were fed routinely and were given equal doses of normal saline. The remaining four groups were inoculated with H22 hepatoma cell lines to prepare a model of hepatocellular carcinoma, and the rats in the model group were given equal doses of normal saline. The high, medium and low doses of Schisandra chinensis polysaccharides were infused 40 mg · kg-1, 20 mg · kg-1 and 10 mg · kg-1 Schisandra chinensis polysaccharides. After 1 week, the vena cava was removed and the serum was separated. The levels of immune cells were measured by flow cytometry. Body, spleen and thymus, calculated tumor inhibition rate, spleen index and thymus index. Results Compared with the control group, the level of serum immune cells and the thymus index in the model group were significantly decreased (P <0.05), but there was no significant difference in the spleen index (P> 0.05). Compared with the model group, (P <0.05 or P <0.01). The increase of serum immunocyte level and thymus index of high dose mice increased significantly with the increase of dose Cell level and thymus index had no significant difference with the control group (P> 0.05). After calculation, the inhibitory rates of Schisandra chinensis polysaccharide in low, medium and high dose groups were 18.7%, 41.1% and 61.4%, respectively. Conclusion Schisandra polysaccharide can improve the cellular immunity and organ immune function of mice at the same time, and thus play a certain tumor inhibitory effect, and the effect shows a typical dose-dependent within a certain range, with the dose increased.
