目的:对中国被毛孢菌丝体(Hirsutella sinensis mycelium,简称HSM)治疗特发性肺纤维化(IPF)大鼠的作用及其作用机制进行研究和探讨。方法:取40只Wistar大鼠分为5组:正常对照组、模型对照组、HSM高、低剂量组(1.0,0.5 g.kg-1)、阳性对照组(氢化可的松10 mg.kg-1)。除对照组大鼠外,大鼠气管注入5 mg.kg-1博莱霉素进行肺纤维化造模,连续给药28 d后,对各组大鼠肺功能、肺组织羟脯氨酸、细胞因子和病理学进行观察。结果:IPF大鼠给予中国被毛孢菌丝体后,HSM 0.5,1.0 g.kg-1能显著降低大鼠肺脏指数和羟脯氨酸含量(P<0.01),同时能明显改善肺功能、肺组织肺泡炎症和肺纤维化程度(P<0.05,P<0.01);1.0 g.kg-1HSM能显著降低大鼠肺组织中TNF-α,IL-1β,TGF-β1蛋白表达(P<0.01),同时能升高肺组织中IFN-γ蛋白表达(P<0.05)。结论:中国被毛孢菌丝体对大鼠肺纤维化有较好的治疗作用,其治疗作用机制与调节TNF-α,IL-1β,TGF-β1和IFN-γ失衡有关。
Objective: To study the role of Hirsutella sinensis mycelium (HSM) in the treatment of idiopathic pulmonary fibrosis (IPF) in rats and its mechanism. Methods: Forty Wistar rats were divided into five groups: normal control group, model control group, HSM high and low dose groups (1.0, 0.5 g.kg-1), positive control group (hydrocortisone 10 mg.kg -1). In addition to the control group rats, rat bleomycin 5 mg.kg-1 intratracheal injection of pulmonary fibrosis modeling, continuous administration of 28 d, the lung function of rats in each group, lung tissue hydroxyproline, Cytokines and pathology were observed. RESULTS: HSM 0.5 and 1.0 g.kg-1 could significantly reduce the lung index and hydroxyproline content (P <0.01) of rats with IPF, but also significantly improve lung function, The levels of TNF-α, IL-1β and TGF-β1 in the lung tissue were significantly decreased in 1.0 g · kg-1HSM group (P <0.01, P <0.01) ), At the same time increase the expression of IFN-γ in lung tissue (P <0.05). CONCLUSION: Mycelium of C.cerevisiae has a good therapeutic effect on pulmonary fibrosis in rats. The mechanism of its therapeutic effect is related to the imbalance of TNF-α, IL-1β, TGF-β1 and IFN-γ.
