我们研究和建立了用高效液相色谱仪反相色谱测定血浆中乙胺碘呋酮的方法,在氯仿溶剂中检出限为10ng,血浆中最低有效测定浓度为0.5μg/ml,平均回收率为98.35%,一日内回收率为98.68%,四日内回收率为98.0l%。乙胺碘呋酮的药物动力学过程属二室模型,其参数为:正常人静注150mg后的平均生物半衰期t_(1/2)(β)为13.75小时;家兔静注30mg/kg后的t_(1/2)(β)为7.17小时。
We studied and established a method for the determination of amiodarone in plasma by reversed-phase high performance liquid chromatography with a detection limit of 10 ng in chloroform and a minimum effective concentration of 0.5 μg / ml in plasma.
