目的:观察固本止咳中药对慢性阻塞性肺疾病(COPD)模型小鼠肺组织中中性粒细胞弹性蛋白酶(NE)表达的影响,探讨固本止咳中药治疗COPD抗炎性反应的作用机制及NE与分泌型免疫球蛋白A(SIg A)的内在联系。方法:采用鼻腔滴入脂多糖加熏香烟的方法建立COPD小鼠模型,分为空白对照组、模型对照组、固本止咳中药组;运用免疫组化方法检测小鼠肺组织中NE表达水平,采用ELISA方法检测小鼠气道SIg A分泌水平。结果:模型对照组肺组织中NE表达气道SIg A分泌水平较空白对照组显著增多(P<0.01,P<0.05),固本止咳中药组较模型对照组显著减少(P<0.01,P<0.05)。结论:固本止咳中药可改善COPD模型小鼠肺组织及气道黏膜炎性浸润状态。
Objective: To observe the effect of Guben Cough Relieving Herbs on the expression of neutrophil elastase (NE) in the lung tissue of chronic obstructive pulmonary disease (COPD) model mice and to explore the mechanism of action of Guben Cough Relieving Herbs on the anti-inflammatory reaction of COPD and its mechanism. The intrinsic link between NE and secretory immunoglobulin A (SIg A). Methods: COPD mice model was established by intranasal instillation of lipopolysaccharide plus cigarette smoke. The mice were divided into blank control group, model control group and Guben Cough Relieving Chinese Herbal Medicine group. The expression of NE in lung tissue was detected by immunohistochemistry, The airway SIg A secretion level in mice was detected by ELISA. Results: The secretion of SIg A in the airway of NE expression in the model control group was significantly higher than that in the blank control group (P <0.01, P <0.05), and the Guben Cough Indication group was significantly lower than the model control group (P <0.01, P < 0.05). Conclusion: Guben Cough Relieving Herbs can improve lung tissue and airway mucosal inflammatory infiltration in COPD model mice.
