目的探讨白藜芦醇(Res)对佐剂性关节炎(AA)模型大鼠关节滑膜组织的抗炎作用和对血管内皮生长因子(VEGF)表达水平的影响。方法 60只SD大鼠随机分正常组(10只)和模型组(50只),模型组左后足趾皮内注射弗氏完全佐剂建立AA模型,模型组在建模后12 d再分5组:模型对照组、Res低剂量组(5 mg/kg)、Res中剂量组(15 mg/kg)、Res高剂量组(30 mg/kg)、阳性药塞来昔布组(5 mg/kg),给药16 d后处死大鼠,对各组大鼠踝关节滑膜组织行病理切片、HE染色和VEGF免疫组化染色。观察滑膜增生、血管翳形成、炎症渗出、关节软骨和骨破坏等情况。Western blot法检测VEGF蛋白的表达情况。结果 Res治疗组的关节病理学评分明显低于模型对照组,免疫组化染色以及Western blot结果显示,Res能降低滑膜组织VEGF表达水平。结论 Res有改善大鼠关节滑膜炎症、抑制滑膜组织VEGF表达的作用,这可能与Res治疗AA关节炎有关。
Objective To investigate the anti-inflammatory effect of resveratrol on synovial tissue of rats with adjuvant arthritis (AA) and its effect on the expression of vascular endothelial growth factor (VEGF). Methods Sixty SD rats were randomly divided into normal group (n = 10) and model group (n = 50). AA model was established by intradermal injection of Freund’s complete adjuvant in model group, Res control group, Res low dose group (5 mg / kg), Res medium dose group (15 mg / kg), Res high dose group (30 mg / / kg). After 16 d of administration, rats were sacrificed and pathological sections of synovial tissue of ankle joints were made in each group. HE staining and VEGF immunohistochemical staining were performed. Synovial hyperplasia, angiogenesis, inflammatory exudation, articular cartilage and bone destruction were observed. Western blot was used to detect the expression of VEGF protein. Results The pathological score of Res group was significantly lower than that of model control group. The results of immunohistochemistry and Western blot showed that Res could decrease the expression of VEGF in synovial tissue. Conclusions Res can improve synovitis and inhibit the expression of VEGF in synovial tissue, which may be related to the treatment of AA with Res.
