目的应用蛋白质组学技术寻找非霍奇金淋巴瘤与健康对照组血清中差异表达的蛋白。方法选择山西省肿瘤医院72例非霍奇金淋巴瘤患者,男性47例,女性25例;年龄11~83岁,中位年龄52.5岁。正常对照组32例,男性17例,女性15例;年龄10~74岁,中位年龄26.5岁,均为健康志愿者。应用SELDI-TOF-MS技术通过WCX-2蛋白芯片检测他们血清中的蛋白质谱,运用Ciphergen Proteinchip 3.0版本的分析软件自动采集数据,结果采用Biomarker Wiz-ard软件分析二组间蛋白质谱中的差异蛋白。结果笔者在非霍奇金淋巴瘤患者的血清中找到了一系列特异表达的蛋白质,其质荷比分别为3193 Da,3398 Da,7974 Da,8564 Da,8693 Da和15934 Da。72例NHL患者与30例正常人的血清中有5个差异明显的潜化标志蛋白,患者血清中高表达且正常人血清低表达的蛋白质的相对分子量为15 934 Da,患者血清中低表达且正常人血清高表达的蛋白质相对分子量为3193 Da,3398 Da,8564 Da,8693 Da。结论质荷比3193 Da,3398 Da,8564 Da,8693 Da和15 934 Da的蛋白质可能是非霍奇金淋巴瘤的生物学标志物。
Objective To find out the differentially expressed proteins in serum of non-Hodgkin’s lymphoma and healthy controls by using proteomics techniques. Methods Totally 72 patients with non-Hodgkin’s lymphoma from Shanxi Cancer Hospital were selected, 47 males and 25 females, aged from 11 to 83 years with a median age of 52.5 years. 32 cases of normal control group, 17 males and 15 females; aged 10 to 74 years, median age 26.5 years old, are healthy volunteers. The protein profiles of their serum were detected by SELDI-TOF-MS using the WCX-2 protein chip. Data were automatically collected using the Ciphergen Proteinchip version 3.0 analysis software. Biomarker Wiz-ard software was used to analyze the differential proteins in the protein spectra . Results The authors found a series of specifically expressed proteins in the serum of non-Hodgkin’s lymphoma patients with mass-to-charge ratios of 3193 Da, 3398 Da, 7974 Da, 8564 Da, 8693 Da and 15934 Da, respectively. There were 5 differentially-expressed marker proteins in sera of 72 NHL patients and 30 normal individuals. The relative molecular weight of the proteins with high expression in serum and low expression in normal human serum was 15 934 Da. The serum level was low and normal The relative molecular weights of the highly expressed human serum were 3193 Da, 3398 Da, 8564 Da, 8693 Da. Conclusion The proteins with mass-to-charge ratios of 3193 Da, 3398 Da, 8564 Da, 8693 Da and 15 934 Da may be biomarkers for non-Hodgkin’s lymphoma.
