目的:研究健脾理气方对人胃癌移植瘤小鼠的5-Fu代谢过程的影响。方法:将80只小鼠按照随机分为人胃癌移植瘤模型组、5-Fu化疗组、健脾理气中药组、5-Fu化疗+中药组,每组20只。将MKN-8荷瘤小鼠新鲜癌块磨碎在小鼠腋窝下接种,制造人胃癌移植瘤小鼠模型。通过高效液相色谱法(HPLC法)检测各组小鼠移植瘤组织及血浆中5-Fu的浓度。结果:中药+化疗组小鼠移植瘤组织中的5-Fu浓度高于单纯化疗组(P<0.05);中药+化疗组与单纯化疗组血浆中的5-Fu浓度则无显著差异(P>0.05)。结论:中药健脾理气方能够提高化疗药物5-Fu在荷瘤小鼠肿瘤组织中的浓度,起到增效的作用。
Objective: To study the effect of Jianpi Liqi prescription on 5-Fu metabolism in human gastric cancer xenografts. Methods: Eighty mice were randomly divided into model group, 5-Fu chemotherapy group, Jianpi Liqi TCM group and 5-Fu chemotherapy + TCM group, with 20 mice in each group. The fresh tumor mass of MKN-8 tumor-bearing mice was ground and ground in the armpits of mice to make a mouse model of human gastric cancer xenografts. The concentration of 5-Fu in the transplanted tumor tissue and plasma of each group was detected by HPLC. Results: The 5-Fu concentration in transplanted tumor tissue of the traditional Chinese medicine + chemotherapy group was higher than that of the chemotherapy alone group (P <0.05). There was no significant difference in the concentration of 5-Fu between the traditional Chinese medicine plus chemotherapy group and the chemotherapy alone group (P> 0.05). Conclusion: The Chinese medicine Jianpi Liqi can increase the concentration of chemotherapeutic 5-Fu in the tumor tissue of tumor-bearing mice and play a synergistic effect.
