目的详细说明建立一种操作简便、稳定可靠的局灶性脑缺血再灌注模型的方法与注意事项。方法 24只雄性SD大鼠随机分两组(模型组和假手术组)。参照并改进Koizumi及Zea-Longa腔内线栓法制备大脑中动脉阻断(MCAO)模型。结果模型组大鼠神经缺陷体征明显,梗死灶明显,梗死部位较恒定;假手术组脑组织结构完整,未见缺血性改变,TTC染色无梗死灶。结论用线栓法制备局灶性脑缺血再灌注模型是成功的。
Objective To describe in detail the establishment of a simple, stable and reliable method of focal cerebral ischemia-reperfusion model and precautions. Methods Twenty-four male SD rats were randomly divided into two groups (model group and sham operation group). The middle cerebral artery occlusion (MCAO) model was made by referring to and improving Koizumi and Zea-Longa intraluminal approach. Results In the model group, the signs of neurological deficit were obvious, the infarct was obvious and the infarct location was relatively constant. The brain tissue of the sham operation group was intact with no ischemic changes and TTC staining without infarction. Conclusion The focal cerebral ischemia-reperfusion model was successfully established by thread-plug method.
