目的探讨甲状旁腺组织中细胞周期素D1(cyclin-D1)及视网膜母细胞瘤易感基因(Rb)的表达及临床意义。方法应用免疫组化技术对36例甲状旁腺腺瘤,10例正常甲状旁腺组织及10例甲状旁腺增生中cyclin-D1及Rb肿瘤抑制基因的表达进行检测。结果腺瘤组cyclin-D1的表达明显高于正常组及增生组(P<0.05),正常组同增生组比较差异无显著性(P>0.05)。Rb的表达正常组、增生组和腺瘤组三者之间比较差异均无显著性(P>0.05)。cyclin-D1过度表达和Rb表达与肿瘤大小、血清钙水平及甲状旁腺素(PTH)水平无关。结论cyclin-D1的过度表达在甲状旁腺腺瘤发生中起重要作用。cyclin-D1过度表达导致Rb磷酸化或表达下调而失去细胞抑制效应可能为甲状旁腺腺瘤发生的机制之一。
Objective To investigate the expression and clinical significance of cyclin D1 (D1) and retinoblastoma (Rb) in parathyroid tissue. Methods The expressions of cyclin-D1 and Rb tumor suppressor genes in 36 cases of parathyroid adenoma, 10 cases of normal parathyroid tissue and 10 cases of parathyroid gland hyperplasia were detected by immunohistochemistry. Results The expression of cyclin-D1 in adenoma group was significantly higher than that in normal group and hyperplastic group (P <0.05). There was no significant difference between normal group and hyperplastic group (P> 0.05). Rb expression in normal group, hyperplasia group and adenoma group were no significant difference between the three (P> 0.05). The overexpression of cyclin-D1 and the expression of Rb were not associated with tumor size, serum calcium level and PTH level. Conclusion The overexpression of cyclin-D1 plays an important role in the pathogenesis of parathyroid adenoma. Overexpression of cyclin-D1 leads to downregulation of Rb phosphorylation or loss of cytostatic effect may be one of the mechanisms of parathyroid adenoma.
