目的:研究马齿苋多糖(POP)对免疫抑制小鼠细胞因子的影响,并探讨其对小鼠免疫功能作用机制。方法:注射环磷酰胺(CTX)建立环磷酰胺诱导免疫抑制小鼠模型,灌胃(ig)给予不同剂量的POP(180和90 mg·kg~(-1))和阳性对照药香菇多糖片(100 mg·kg~(-1)),通过计算小鼠模型前后脾脏指数和胸腺指数。MTT法测定淋巴细胞增殖反应,双抗夹心ELISA法测定小鼠血清中细胞因子IL-2,IL-4,IL-10,IFN-γ,NK和LAK的含量。结果:POP可增强CTX所致小鼠自然杀伤细胞和淋巴因子激活的杀伤细胞活性,并能增加血清细胞因子IL-2,IL-4,IL-10和IFN-γ的含量;POP可提高免疫抑制小鼠的脾脏指数和胸腺指数。结论:马齿苋多糖能明显提高小鼠机体的免疫能力,恢复由CTX诱导的小鼠免疫抑制状态,这可能是其抗肿瘤活性的作用机制之一。
OBJECTIVE: To study the effect of the polysaccharide of portulaca oleracea (POP) on cytokines in immunosuppressed mice and its mechanism of immune function in mice. Methods: Cyclophosphamide-induced immunosuppressive mice model was established by injection of cyclophosphamide (CTX). Different doses of POP (180 and 90 mg · kg -1) (100 mg · kg -1), the spleen index and thymus index were calculated before and after the mouse model. The proliferation of lymphocytes was measured by MTT assay. The levels of IL-2, IL-4, IL-10, IFN-γ, NK and LAK in serum were measured by double antibody sandwich ELISA. Results: POP enhanced CTX-induced natural killer and lymphokine-activated killer cell activity and increased serum levels of cytokines IL-2, IL-4, IL-10 and IFN- Inhibition of mouse spleen index and thymus index. Conclusion: Polysaccharide from Portulaca oleracea can significantly enhance the immune capacity of mice and restore the immunosuppressive state of mice induced by CTX, which may be one of the mechanisms of antitumor activity.
