目的:研究脂肪酸-辅酶A连接酶4(FACL4)在胃癌中的表达,及其与8-溴-环腺苷酸(8-Br-cAMP)调控胃癌细胞增殖、凋亡的关系。方法:采用逆转录聚合酶链反应技术,检测23例胃腺癌组织和癌旁正常胃黏膜、以及8-Br-cAMP作用SGC7901细胞后FACL4 mRNA的表达水平。应用流式细胞仪及透射电镜技术,检测2×10-5mol/L浓度的8-Br-cAMP作用48 h后SGC7901细胞的凋亡情况。结果:与正常胃黏膜相比,胃腺癌组织及SGC7901细胞中FACL4 mRNA表达水平显著上调(P<0.05),胃腺癌组织FACL4 mRNA阳性表达率为87%。2×10-5mol/L的8-Br-cAMP可抑制SGC7901细胞中FACL4的表达,从而抑制胃癌细胞增殖,诱导细胞凋亡。结论:FACL4在胃癌组织中表达增高,8-Br-cAMP作为选择性FACL4抑制剂,可抑制胃癌细胞增殖和FACL4表达,诱导胃癌细胞凋亡,可能成为胃癌诊治的一个新靶点。
OBJECTIVE: To study the expression of FACL4 in gastric cancer and its relationship with the regulation of 8-Br-cAMP on the proliferation and apoptosis of gastric cancer cells. Methods: Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of FACL4 mRNA in 23 cases of gastric adenocarcinoma and adjacent normal gastric mucosa, and 8-Br-cAMP in SGC7901 cells. Flow cytometry and transmission electron microscopy were used to detect the apoptosis of SGC7901 cells treated with 8-Br-cAMP at 2 × 10-5mol / L for 48 h. Results: Compared with normal gastric mucosa, the expression of FACL4 mRNA in gastric adenocarcinoma and SGC7901 cells was significantly up-regulated (P <0.05). The positive expression rate of FACL4 mRNA in gastric adenocarcinoma was 87%. 2 × 10-5mol / L 8-Br-cAMP can inhibit the expression of FACL4 in SGC7901 cells, thereby inhibiting the proliferation of gastric cancer cells and inducing apoptosis. CONCLUSION: FACL4 is highly expressed in gastric cancer. 8-Br-cAMP, a selective inhibitor of FACL4, can inhibit gastric cancer cell proliferation and FACL4 expression and induce gastric cancer cell apoptosis, which may be a new target for the diagnosis and treatment of gastric cancer.
