黄芪预处理对缺血再灌注大鼠心肌细胞凋亡及相关基因的影响(英文)

背景:黄芪具有保护缺血再灌注细胞损伤的作用,黄芪预处理是否对缺血再灌注心肌细胞凋亡有保护作用?目的:探讨黄芪预处理对缺血再灌注大鼠心肌细胞凋亡及其相关基因的影响。设计:以Wistar实验大鼠为实验对象的随机分组对照。单位:承德医学院基础医学部及附属医院老年病科。材料:实验于2004-02/12在承德医学院基础医学研究所免疫组化实验室完成。选用雄性Wistar大鼠30只,将动物随机分为黄芪预处理组(黄芪组)、缺血再灌注组、假手术对照组(对照组),每组10只。方法:黄芪组术前给予腹腔内注射黄芪注射液,缺血再灌注组、对照组术前给予等量生理盐水注射。1周后制备缺血再灌注模型,术后即取缺血再灌注边缘区的心肌,对照组取相对应部位心肌。应用脱氧尿嘧啶缺口末端标记法测定心肌凋亡细胞指数的改变,应用免疫组化ABC法检测心肌细胞bcl-2(抑凋亡基因)及bax(促凋亡基因)基因的改变。主要观察指标:①各组凋亡细胞数。②bcl-2和bax基因表达情况。结果:①心肌凋亡细胞数:黄芪组低于缺血再灌注组[(14.06±9.97)%,(19.34±12.30)%,t=1.863,P<0.05]。②bcL-2基因表达:黄芪组与缺血再灌注组无明显差异[(9.14±4.46)%,(8.99±4.54)%,P>0.05]。③bax基因表达:黄芪组低于缺血再灌注组[(12.65±7.23)%,(18.12±7.92)%,t=2.096,P<0.05]。结论:黄芪预处理可使促凋亡基因bax的表达明显下调,从而使心肌细胞凋亡减少,保护缺血再灌注心肌细胞。 BACKGROUND: Astragalus has a protective effect on ischemic reperfusion injury, and whether astragalus preconditioning protects cardiomyocytes against ischemia-reperfusion injury. Purpose: To investigate the effect of astragalus preconditioning on myocardial cell apoptosis induced by ischemia-reperfusion in rats. The effect of related genes. Design: Randomized grouping of Wistar rats as experimental subjects. Unit: Department of Geriatrics, Basic Medicine Department, Affiliated Hospital, Chengde Medical College. MATERIALS: The experiment was performed at the Laboratory of Immunohistochemistry, Institute of Basic Medicine, Chengde Medical College from February to December 2004. Thirty male Wistar rats were used. The animals were randomly divided into astragalus pretreatment group (Huangqi group), ischemia-reperfusion group, and sham-operated control group (control group), with 10 rats in each group. Methods: Astragalus group was given intraperitoneal injection of astragalus injection before operation, ischemia and reperfusion group and control group were given normal saline injection before operation. One week later, the ischemia-reperfusion model was prepared. After the operation, the myocardium in the marginal region of ischemia-reperfusion was taken. In the control group, the corresponding part of the myocardium was taken. Apoptotic cell index was determined by deoxyuridine nick end labeling. The changes of bcl-2 (apoptotic gene) and bax (proapoptotic gene) were detected by immunohistochemical ABC method. MAIN OUTCOME MEASURES: 1 Number of apoptotic cells in each group. 2bcl-2 and bax gene expression. RESULTS: 1 The number of apoptotic myocytes was lower in the Astragalus group than in the ischemia-reperfusion group [(14.06±9.97)%, (19.34±12.30)%, t=1.863, P<0.05]. 2bcL-2 gene expression: There was no significant difference between the Astragalus group and the ischemia-reperfusion group [(9.14±4.46)%, (8.99±4.54)%, P>0.05]. The 3bax gene expression was lower in the Astragalus group than in the ischemia-reperfusion group [(12.65±7.23)%, (18.12±7.92)%, t=2.096, P<0.05]. Conclusion: Astragalus membranaceus pretreatment can significantly reduce the expression of the pro-apoptosis gene bax, so as to reduce the apoptosis of cardiomyocytes and protect myocardial cells from ischemia-reperfusion.
